4. Explain the procedure producing pain and/or distress.
Thirty-one- rhesus monkeys were used as part of our program to evaluate
drug dependence liability of new compounds. These rhesus monkeys are
physically dependent on morphine and periodically allowed to go into
spontaneous withdrawal or withdrawal is precipitated by opiod antagonists.
During this time they undergo moderate to severe stress. On completion
of the observatiod period, withdrawal signs and symptoms are relieved by
an injection of morphine. It is important to note that these experiments are
repetitive in nature. A new investigational chemical is being tested on
each occasion. Many of these chemicals are opiod in nature and prevent
the appearance of the withdrawal signs and symptoms.

The distress produced by this procedure is the end-point being measured.
• As indicated above, many of the compounds tested relieve these symptoms
and the experiment is terminated immediately after the observation

This laboratory uses a well-defined guinea pig- model of gallbladder inflama-
tion (acute cholecystitis) to evaluate the mechanisms that initiate acute
inf lamation.

Since analgesics inhibit inf lamation as part of their action, they wpuld
interfere with the study results. Most animals get analgesics/ anti i rift amatory
drugs as part of the experimental protocol, but about 30 % do not.
